Fisetin to Clear Senescent Cells

Following studies with mice that showed significant senolytic clearance of senescent cells following large doses of the readily available flavenoid supplement Fisetin,  my wife and I (ages 79 and 84) decided to try it.  We have just completed two sets of massive Fisetin doses.

We had Life Extension blood-work done in October before the start, and we will have more again next week to observe any changes.  The first set of Fisetin doses was on October 22-25 with 800 mg/day for three days followed by 600 mg on the fourth day, for a total of 4 g.  I didn't notice much in the way of effects.  Perhaps some reduction of small aches and pains and some increase in energy and mental acuity.

For the second set of doses done November 22-26, since we experienced no negative side effects in the first set we decided to increase the dosage a bit and to add 10 mg of BioPerine, a supplement that is reputed to magnify the effects and potency of flavenoids.  For five days starting on Thanksgiving we took 500 mg of Fisetin and 10 mg of BioPerine twice per day, for a total of 5 g of Fisetin.

This time. I did experience one negative side effect.  A few months ago, about 2 AM in the morning I awoke from a deep sleep and experienced a severe episode of vertigo.   I turned over in bed, and the the whole room seemed to tilt.  Suddenly, I didn't know which way was up.  I staggered to the bathroom and vomited.  The symptoms tapered off and disappeared in a few days, but it was a very distributing experience.

On the 2nd day of our 2nd Fisetin series, I experience a recurrence of that vertigo in the middle of the night, not as bad as my initial experience but still rather disturbing.  I tolerated this mild vertigo and continued the treatment.  My wife had no similar symptoms, and after my last dose I experienced no further vertigo symptoms.

On the positive side, following the second set of dosages I did feel very well, and very sharp and alert.  This past weekend I ran my Shetland Sheepdog Taliesin in an AKC Canine Agility Trial in Mt. Vernon, WA, and we did very well, qualifying in 7 runs out of 15 and getting various colored placement ribbons.  I was feeling quite sharp, and I even invented a new dog-handling technique that fixed an ongoing problem we were having.

Next week we will do the blood-work again, and I'll report any changes.

  • My 41 yr young wife just took her first 2 capsules of " updated w fisetin "life extension " senolytics " caps with grapefruit juice on a relatively empty stomach followed by raking/ weeding in the  yard....   she is now experiencing energetic,  euphoric feelings.   She will take this 3 days in a row...   last time she tried this approach, " without fisetin" she looked 18 yrs old!!   Anyone else had similar experiences?

      Hi to all,

      I am a 47 year old female pharmacist (retired to run an education business) in Australia weighing 59kg. No current health problems. Latest bloods - all within normal limits.

      I have just finished my 4th fisetin trial over 5 days.

      I do a fisetin trial every 4-6 months using fisetin from Vitaspace (powder form). Vitaspace provide a spoon which measures 620mg.

       

      Latest dosing as follows:

       

      Day 1: 3100mg (620mg x 5 Vitaspace spoonfuls) - dissolve and mix thoroughly in equal parts of Polysorbate 80, extra virgin olive oil and hot water with a dissolved 1/2 Equal artificial sweetener tablet. It becomes a very smooth toothpaste consistency. Eat with a spoon. Rinse with strong coffee with real cream (this is the cream you get from the dairy section and might whip to put on top of a meringue cake).

      Do not eat anything for 30 minutes.

       

      Day 2: 3100mg (620mg x 5 Vitaspace spoonfuls)

      Day 3: 3720mg (620mg x 6 Vitaspace spoonfuls)

      Day 4: 4340mg (620mg x 7 Vitaspace spoonfuls)

      Day 5: 4960mg (620mg x 8 Vitaspace spoonfuls)

       

      Results:

      Day 1: nothing, had a nap after lunch

      Day 2: 90% reversal of chronic nasal congestion, 50 - 60% more energy, faster urination.

      Day 3: woke with more than normal amount of energy

      Day 4: nothing of note

      Day 5: Got COVID-19. Runny nose lasted for three days. Used my Ventolin puffer a few times for a dry cough which became a wet cough for 1-2 days after the runny nose finished.

    I tried it again...1,300 mg same brand fisetin with added black pepper, and the quercetin. Again, nothing to report. I will use Swanson next time.

    • Dale Piggott To Moonlitnight and all others who have mentioned (or take) quercetin. www.Mountsinai.org states in part that: " Anticoagulants (blood thinners). Quercetin may enhance the effect of these drugs, increasing your risk for bleeding. Anticoagulants include:."  So, consider it as well..

    Also, how often is this repeated (on average) by the experimenters here?   I can't seem to find any references other than Fairy's, which I am presently planning to follow. However, Fairy is not in her very late '60s like I am. Thank you.

    I'm bumping this fantastic thread. I know the Mayo Clinic trial's dosage is Fisetin 20mg/kg/day, orally for 2 consecutive days. And I understand we repeat this dosage within 2 to 3 months.  My question is: when do we repeat the two month protocol? In six months? In one year? Or never?  Thanks!

      ripeideas The truth is that the researchers at the Mayo Clinic are guessing!  The two consecutive days is based on limited murine studies, but in a longer lived mammal like humans, one could argue for a longer period.  The researchers at the Life Extension Foundation are suggesting chronic use of senolytics on a once weekly basis.

      ripeideas I read a study on Quercitin showing that it does not directly induce apoptosis but opens several pathways for apoptosis, presumably ripe for a senolytic like fisetin.  Seems like it should be considered, esp liposomal.

      ripeideas I don't think anyone really knows yet what frequency is best.  One idea is to base it on how long it takes for senescent cells to accumulate after a treatment.  A month or so I think, but will depend on age and health.

      Rob8311 I wish I didn't get so damned depressed on the choline content of liposomal supplements. I actually made my own liposomal Vitamin C and resveratrol but I was depressed as all get out. Strange. Melatonin does the same thing. I'm not a depressive person!

      Rob8311  Lack of a practical means for measuring the percentage or volume of senescent cells in the body seems to be the biggest problem with determining the effectiveness of any senolytic treatment.    Autopsy seems to be the only reliable method and has been used in tests involving lab mice but is obviously impractical for human testing.    I keep hoping someone will come up with a blood or urine test which will provide a relative indication of the volume or percentage of senescent cells in a human body or an indication of cells being destroyed by in vivo following a senolytic treatment.    Perhaps some residue from ruptured senescent cells released into the bloodstream or excreted in urine.....

      ripeideas That sucks.  I have my own insomnia problem, not the same as yours I know.  THC/CBN gummies worked great, for a while.  Sounds like you are using an overall health approach instead of playing whack-a-mole with symptoms.  Having said that, have you tried LDN?

      Rob8311 Just ordered a test from The DNA Company (damn webinars!).  Maybe I'll be able to see if I am somehow triggering the wrong gene.  Not an endorsement because I haven't tried it yet, just brainstorming.  I didn't react to LDN, but I have tried that and other interventions through AgelessRX.

    Just wanted to share this older study regarding the reduction of Fisetin methylation (into less effective geraldol) in the liver when combined with increasing amounts of red wine:

  • Hi Everyone!  I just stumbled across this thread and find it very informative. I would like to ask the group their opinion of what I should do next.  I am 65 and have had chronic pain for 10 years: knee pain, L4-5 issues, nerve pain in butt (probably die to L4/5), neck pain (severe OA there) and more recently should/arm pain. It just keeps expanding to new areas. I work with an Anti-Aging institute and they can find little to work with as my blood work is, in their words, impressive: lipid panel 30 yr olds would love to have (their words), C reactive protein WNL, nutrients all good, liver enzymes low, etc...I feel like I have tried everything with no symptom relief.  Here is what I've done over the last couple years:  a variety of Peptides ( one combination worked on my knees but we couldn't replicate it), PPS, EGCG, high doses of Melatonin, bioidentical hormone replacement, intermittent fasting, Spermidine, and a stint of D+Q about a year ago; all of this they recommended to me to reduce inflammation (which hasn't worked). I thought maybe I was focusing on the wrong thing so I took a blood test that measures cytokines and I'm waiting for the results.  After reading much of this thread, I decided that my D+Q protocol was under-dosed, so just yesterday I started a more aggressive protocol of 200 mg D+ 1250mg of Quercetin which I will do for 4 days. Does anyone have any other ideas of what else I can try? I would really appreciate your suggestions. Thank you in advance.

    • Denise Shugerman Have you tried high dose fish oil? Over the counter is cheap and you can get analysis of various brands if you are member of Consumer Labs. To get about 5 grams of EPA/DHA costs about $15/month at Costco.  

      A quick check on Google will document the many benefits of high dose fish oil for most of your issues ( and many more). You might want to check out one of Barry Sears books.

      Peter H. Howe I use to be a member of Consumer Labs.  The big problem they have is you as a supplement manufacture/distributor must pay them to test your product.  They will not test anyone/s product that does not pay.  So they charge the consumer and the supplier. Many best products are never tested. 

        In my mind a BAD WAY TO DO BUSINESS.  I do not trust them

    • Van Thank you for the information. I was not aware of the conflict and it is a conflict. May be the reason why my fish oil is no longer on the list and earlier tests are no longer mentioned. I have, however,  seen occasions when they have reported different conclusions on two different products from the same supplier.
