A Senolytic Alternative: D+F+P?

It has been suggested to me that an alternative to the now-standard D+Q senolytic treatment might be a D+F treatment, i.e., substituting Fisetin for Quercetin.  The two chemical compounds are both flavenoids, and the Neiderhoffer paper shows that Fisetin is better at killing senescent cells than is Quercetin.  So it seems to be a reasonable surmise that Fisetin might work even better in combination with Dasatinib.

Further, one might add 10 mg of BioPerine, since the compound Piperine is known to suppress the production of P-glycoprotein,  a protein that the body uses to break down exogenous compounds found in the body and that leads to the short half-lives for flavenoids in the blood stream.  (My own experience with large Fisetin doses is that when 10 mg of BioPerine was added, I experienced a mild temporary vertigo side-effects, which I take as indicating a magnification of the Fisetin action.)

Does anyone have any comments about this D+F+P proposal?  Any thoughts about dose sizes?

  • I found the easiest and best forurce of piperine is dried indian long peppers that are easily found in packaged dehydrated form in any Indian grocery store. I find it is best if you put about 10 dried long peppers to soak in a small glass of water about half an inch deep and then after soaking 1 to two hours thay are easy to either chew or swallow. If you try to chew the dried form they make your tongue sort of numb.

    We just did D+Q+F+P.

    On careful consideration, after having done two rounds of D+Q+P a week apart, my wife and I decided to finish off the series a week later by adding Fisetin and doing a round of D+Q+R+P.  This morning at 8:30 AM we took 10 mg of Bioperine, intended to suppress the action of drug-removing P-glycoprotein before the other molecules enter the bloodstream.   At 9:30 AM we took 100 mg of Dasatinib (2 tablets) + 1,200 mg of Quercetin (1 capsule) + 1,000 mg of Fisetin (10 capsules).  Then we had breakfast.  At 11:30 AM we took another 10 mg of Bioperine.  At 12:30 PM we again took 100 mg of Dasatinib (2 tablets) + 1,200 mg of Quercetin (1 capsule) + 1,000 mg of Fisetin (10 capsules).  Then we had lunch and watched Colts vs. Texans.  My distant cousin Andrew Luck did very well in the game.

    it's now 5:00 PM.  So far no side-effect symptoms.  In the previous D+Q+P sessions I had experienced cramps, mild diarrhea, a mild headache, and a general feeling of unwellness, but so far I have experienced nothing like that this time.  I'll report any later developments.
