Fisetin ( + quercitin)

Among flavonoids, fisetin appears to be an effective senolytic:

https://www.fightaging.org/archives/2018/10/animal-data-shows-fisetin-to-be-a-surprisingly-effective-senolytic/

"Fisetin appears about as effective in mice as any of the current top senolytics, such as the chemotherapeutics dasatinib and navitoclax."

Because it is cheaper, easier to acquire, and probably safer than dasatinib, self-experimenters should consider examining fisetin in combination with quercitin.

  • Effects of Fisetin dosing over 6 months.

Now in my 5th round of Fisetin dosing over the past 6 months.  Some noticeable effects now that were not apparent in the early rounds.  Examples below.

I cannot say all effects are direct result of Fisetin, but effects occur immediately on starting a Fisetin round, and then taper off until the next round.

    - Improved balance.  At age 76 more aware of balance than in younger years.   Fisetin seems to improve it.  Example: In front of my apartment building are 4 steps, no handrail.  So I used to carefully position myself next to a post and hold on before stepping down.  After taking a round of Fisetin, no need to do that, just step down.  Surprised me.  Is it possible Fisetin is the cause of that change?

    - Improved mental function.  My main mental challenge these days is learning a foreign language; an extremely difficult, Oriental language (Thai).  Each time I take a round of Fisetin there is noticeable improvement in my listening comprehension and reading speed.    Modest, but noticeable and very welcome.

    - Sharper vision.  My vision has been steady for many years, but after recent rounds of Fisetin noticeably brighter and sharper.   I’ve even gone without glasses a few times.

    - Easier urination.  An important function for old men.  Flow is certainly greater after starting a Fisetin dose.

    - Better sleep, but need to sleep and nap a lot more during the 3 days that I take Fisetin.

    My regimen:

    • Fisetin (Doctor's Best brand),  500 mg. per dose (5 capsules of 100 mg. each) 2x/day, for 3 days: 6 doses for total of 3,000 mg.
    • Olive Leaf Extract: 1 capsule
    • Bioperine: 10 mg.
    • Quercetin: 1,000 mg (2 capsules, 500 mg. each)
    • MCT oil: 3,000 mg. (3 capsules, 1,000 mg. each)
    • Coconut oil: 1 Tablespoon
    • (no DMSO, although would be easy to take along with coconut oil)

Back when first starting Fisetin, I diligently emptied all capsules and mixed with coconut oil before swallowing.  But old age is motivating me to simplify things.  Now I just swallow the lot of capsules with water or juice.

I'm very interested in comments, critique and others' experiences over several months of Fisetin dosing.

  • In contrast to my personal observations about Fisetin just above, here is a more rigorous discussion of effects:

I have been taking 1200mg of fisetin in a single dose for 2 days every month. For the last 2 months, I have taken it every 2 weeks because I have liked the effects I have seen and want to see if more frequent doses will increase improvements. I weigh 125 pounds.

The first noticable improvement was not my own, but my mom's. She has had a nickel size flat mole on her cheek for about 15 years. After 2 monthly sessions, it had disappeared and only a light brown freckle the size of a pencil eraser head remains. She has not done more regular sessions. Because of her immediate significant improvement, I started watching my own moles. Ones that have darkened with age have all lightened and decreased in size, though none of mine have disappeared, but I have had them since I was a child where my mom's came in her 50s.

I have injury onset arthritis in my large toe, and the pain has almost completely gone away. While my toe has slowly recovered, the arthritis kept me even from walking more than 30 minutes at a moderate pace without noticable pain for years.

When I take the fisetin, I feel extra tired for a few days and sometimes random aches and pains or muscle soarness, but it goes away after a few days. My husband felt like he had a sunburn the first time he took it (he is fair skinned and burns easily and sometimes blisters). I attribute the pains and soarness to apoptosis and treat it like I do muscle soreness from exercise- in that I try to eat plenty of protein and nutrients to grow new cells and give my body some recovery time.

I am not currently adding in anything to enhance bioavailability because I am seeing impressive results with just the supplement (Swanson brand). Also, at least the high dose study in rats was straight oral gavage, so I don't feel the need to alter treatment.

As a side note, other supplements I have taken for about 4 years: I take 1T of omega3 fish oil daily (4500mg), NAD+ in the form of 2 Elysium pills a day, I have previously taken Mitoq, but haven't for about 4 months, 4000mg VitD, 1000mg VitC, Children's chewable multivitamin, and a probiotic.

I also fast for 18-24 hours once a month from both food and water.

All improvements listed above were after starting Fisetin supplementation about 9 months ago. I would need to do a little more digging for the exact time.

Thank you for your report. It is interesting to learn about your positive experiences from Fisetin. Especially since you first have used a program with a monthly treatment interval and later have had treatments twice monthly. It is fascinating that old moles can disappear after a few treatments.

This might make the rest of us consider more regular treatment intervals  – similar to what you and Mike have done. Personally, I have taken a rest from senolytic treatment with fisetin. This since I did not want to overdo it. Personally, I have incorporated 100 mg fisetin an daily basis.

Staffan Olsson Thanks for your feedback. Daily low dose has been shown to do good things for people, but doesn't necessarily give the senolytic effects. Looking at the mouse study published last year, reduction in senescent cells happened after continual high dosing for 2 weeks or a single very high dose, but once treatment was stopped, senescent cells continued to increase, so regular dosing is needed if you want to keep senescent cells cleared. I decided on monthly because one of the human studies is currently doing a high dose for 2 days a month for 2 months on 70+ year old women. I figured if they could do that, then it was safe for me to do it. I have just continued monthly dosing for longer. I have increased to every 2 weeks because I want to see if it will do even more. I am not worried about toxicity because I am not taking it daily, so my body has time to clear it and recover. Even daily high doses were helpful for 2 weeks in mice. My moles are also a good way for me to monitor effects, so that has given me confidence in increasing frequency of dose. If nothing else, it is possibly reducing the likelihood of skin cancer.

Hello everyone I wanted to make my first post here as I have been doing a fisetin routine for a year now just about and I have results that are not placebo and very profound. I honestly do not know if this is the senylotic effect or some kind of interaction with fisetin in other ways.

To get it out of the way, I will give the fast and short version. I am taking Swanson’s Fisetin. I take around 2g a day for 3 days( now 2.5g). I repeat this for 3 or 4 times with varying intervals (I was trying to find optimal intervals).

My my first regimen was 3 days treatment with a 2 week delay, then a 3 week delay, then a 4 week delay, then a 2 month delay. I am now convinced a 3 week delay is the minimal amount of time you want taking this the way I am, though I will continue with 4 week delay for safety sake and because of my own observations.

So results. Well I feel like this must play on hormone levels in the short term. Immediately after starting a 3 day regimen, usually on day 2 or 3 I feel strong as hell. In particular my legs are tight and super strong. My muscles in general are firmer and overall strength is amazing. My mind is clear, thinking concise and dynamic. I feel more dominant, self confident, and aggressive. Libido is enhanced as well. This all led me to believe that this acts as a testosterone mimic or elevates testosterone in the blood which is why I cycle monthly and then take a few months off every so often.

i can tell you that at the age of 43 this makes me legitimately remember what early 20s feels like. The effects of one 3 day regimen last strongly for 2 weeks and even on for 4 weeks. In particular during week one and two I was amazed at the amount of pure emotion and clarity of the world around me. I could hear and appreciate sounds and colors seemed more vibrant. Mentally I am on overdrive with super memory and crystal clear logical thinking. I remember the first time I took this I was shocked. I was in a cafe just looking at a coke I had bought in a glass amazed at how more real and present it looked. A small thing but it was profound just how the world seemed more alive. It literally felt like going back in time 20 years.

So for long term effects. A stronger posture, better muscle tone, improved gum health, vastly more energy, in fact this has become all the norm after around 8 months of this. Many of the effects I described in the first two weeks following a 3 day regimen do taper off to an extent, but never completely.

As far as side effects there has not been anything thus far. When you are taking the 3 day dose you might get amped up a little jittery but that gives way quickly after the last dosage and it normalizes into a even effect that is positive in nature. I was very concerned about hair loss as I could not believe how impactful this was. But I have noticed no hair loss at all. I cycle it with a 3 or 4 week break to ensure that there is no negative effect on hair or otherwise. With a two week interval I had some scalp itchiness. That was enough to back me off to to four week intervals.

I honestly think that most of what I experienced is not the senylotic effect. It’s way too fast and profound. But I have no ill effects and feel incredible. And overall I have a youthful vigor and look and feel younger. I think a senylotic evaluation, as far as a laymen just observing himself goes ,can only really be done after 2 years or so. However the initial results ( especially gum health) and from what else I can observe are overwhelmingly positive.

I can tell you that this does have an affect and it is profound and noticeable. How much of this is hormone I do not know. But I have other improved health as well, however it’s always tricky to quantify this objectively yourself sometimes.

Good luck on your experiments. Let me know if this has the same effect for you. I just bought Swanson’s with nouvestein. Nothing fancy. Cheap and on amazon.

Also, on vigorous effect to lower my omega 6/ omega 3 ratio to reduce general tendency to inflammation. Eliminated all seed oils & taking 1 gm DHA/EPA supplement BID, so my eczema remission may not be solely due to the hi fisetin intake  Allen DeLaney
