Fisetin experimentation one year (reposted as topic for info purposes)

Hello everyone I wanted to make my first post here as I have been doing a fisetin routine for a year now just about and I have results that are not placebo and very profound. I honestly do not know if this is the senylotic effect or some kind of interaction with fisetin in other ways.

To get it out of the way, I will give the fast and short version. I am taking Swanson’s Fisetin. I take around 2g a day for 3 days( now 2.5g). I repeat this for 3 or 4 times with varying intervals (I was trying to find optimal intervals).

My my first regimen was 3 days treatment with a 2 week delay, then a 3 week delay, then a 4 week delay, then a 2 month delay. I am now convinced a 3 week delay is the minimal amount of time you want taking this the way I am, though I will continue with 4 week delay for safety sake and because of my own observations.

So results. Well I feel like this must play on hormone levels in the short term. Immediately after starting a 3 day regimen, usually on day 2 or 3 I feel strong as hell. In particular my legs are tight and super strong. My muscles in general are firmer and overall strength is amazing. My mind is clear, thinking concise anddynamic. I feel more dominant, self confident, and aggressive. Libido is enhanced as well. This all led me to believe that this acts as a testosterone mimic or elevates testosterone in the blood which is why I cycle monthly and then take a few months off every so often.

i can tell you that at the age of 43 this makes me legitimately remember what early 20s feels like. The effects of one 3 day regimen last strongly for 2 weeks and even on for 4 weeks. In particular during week one and two I was amazed at the amount of pure emotion and clarity of the world around me. I could hear and appreciate sounds and colors seemed more vibrant. Mentally I am on overdrive with super memory and crystal clear logical thinking. I remember the first time I took this I was shocked. I was in a cafe just looking at a coke I had bought in a glass amazed at how more real and present it looked. A small thing but it was profound just how the world seemed more alive. It literally felt like going back in time 20 years.

So for long term effects. A stronger posture, better muscle tone, improved gum health, vastly more energy, in fact this has become all the norm after around 8 months of this. Many of the effects I described in the first two weeks following a 3 day regimen do taper off to an extent, but never completely.

As far as side effects there has not been anything thus far. When you are taking the 3 day dose you might get amped up a little jittery but that gives way quickly after the last dosage and it normalizes into a even effect that is positive in nature. I was very concerned about hair loss as I could not believe how impactful this was. But I have noticed no hair loss at all. I cycle it with a 3 or 4 week break to ensure that there is no negative effect on hair or otherwise. With a two week interval I had some scalp itchiness. That was enough to back me off to to four week intervals.

I honestly think that most of what I experienced is not the senylotic effect. It’s way too fast and profound. But I have no ill effects and feel incredible. And overall I have a youthful vigor and look and feel younger. I think a senylotic evaluation, as far as a laymen just observing himself goes ,can only really be done after 2 years or so. However the initial results ( especially gum health) and from what else I can observe are overwhelmingly positive.

I can tell you that this does have an affect and it is profound and noticeable. How much of this is hormone I do not know. But I have other improved health as well, however it’s always tricky to quantify this objectively yourself sometimes.

Good luck on your experiments. Let me know if this has the same effect for you. I just bought Swanson’s with nouvestein. Nothing fancy. Cheap and on amazon.

One dose. I just take 30 pills one after another drinking water. My reasoning is you want to overload the body’s ability to metabolize it and get it circulating in the blood to affect as many senescent cells as possible.

The half life on this stuff even with large doses is like 3.1 hours, so it should be out of your system effectively in around 9 hours.

If you do take it this way, the first place I notice an effect is the outer thighs. It gets tight and might twitch a little. Leg strength is something I noticed first as well. Drink plenty of water, I used to get some head aches that were not painful just uncomfortable, but that does not happen anymore.

David C Thanks!  Been taking NMN for awhile :-) So I dosed my last dose this morning, I'll report back here on the next few days.  I am not nearly as tired today as I was yesterday.  Also, I would assume to not exercise/lift on the week of doing this, correct?  Not like I really can anyways with the quarantine.  I'll probably try this again in 3-4 weeks but with olive oil or MCT oil just to see.  Cheers!

Chris I lift while taking. I used to not do so on the days I dosed. But i figured I’d try it and see. I noticed I might get more muscle soreness but other than that seems fine. I mean I guess the concern is that fisetin might eliminate those transient senescent cells that are created to attract immune cells for wound healing. Which I guess is the mechanism by which the muscle rebuilds after a workout.

Seemed to me not to be harmful at all to workout though. I feel like I get a good rip to be honest with it.

    • David C I tried your experiment taking 3 g per day for three days of a liposomal fisetin 600mg capsules. I did not notice anything different in how I felt. It could be because I exercise every day,  regularly do fasting and  I am very health conscious but when I did my swimming 20 lap routine over 30 minutes on the second day, I was one minute faster over 20 laps, which is an amazing improvement. Again on the 4th day. I was again another minute faster. So though I didn’t feel any different, when it came to aerobic swimming exercise, I was significantly stronger and faster but it did not show up on my interval training on my bicycle.

I will repeat in three weeks and see if I have more results.

John Mcgough Thanks for sharing your results, John.  I likewise don't notice any difference in how I feel after my monthly fisetin (along with quercetin and theaflavins) protocols, nor did I see the amazing improvement in any of my daily exercise routines that you experienced.  Nonetheless, I certainly will continue this episodic protocol, as I don't expect that a reduction in my rate of aging (or so I hope!) by senolysis would necessarily be noticeable.

    • Brian Valerie I swam again today and amazingly I was another 30 seconds faster. Maybe I am evaluating my interval training on the bicycle wrong. I always try to raise my heart rate to 150 over 20 minutes. I haven’t been successful lately and maybe it’s because my legs are so much stronger it’s easier for me so I’m stuck in the 120s for a pulse rate.

      BTW, I am 74 years old weigh 175,  5’,8”

I’ll see how long this  improvement in swimming laps last? I also take Senolytic  activator by life extensions once a week with my rapamycin dose, plus 2 bio Fisetin daily.

John Mcgough Keep us posted.  My resting heart rate has gradually been going down (to about 55-65), but I expect that's due to my daily exercise rather than senolytics.  We're truly on the frontier of relevant dosing protocol research, but I've opted for tripling Life Extension's weekly dose, taking it only once monthly, Mayo Clinic style.  No negative side effects thus far (of which I'm aware).

I get different effects from different brands of Fisetin supplements. I get a strong effect when I use Dr's best fisetin 30mg/Kg body weight mixed with oliveoil and taken 1 hour after I have taken black pepper/piperine. The very same day I get a minor euphoria and enhanced energy, then decreased performance capacity for a 1 - 2 days.  After 3- 6 days, my endurance capacity is increased.  My joint pain is reduced almost immediately on day one or two.

When I try other products, I get a different effect. I have used life extension's Fisetin with enhanced absorption. It makes me feel weak for a few days after ingestion. I speculate that this happens because of autophagy. But the fisetin from Dr's best bring me other effects that I very much welcome.

Nowadays, I use both, this since they definitely have different effects on my body.

I don't say that Life extension's fisetin in't working. It definitely is. A 88-year-old female family member tried it. And got a strong effect that lasted a few days. She felt very weak, was exhausted and was confused.

      Staffan Olsson

That really is intriguing, Staffan.  It does seem that there is a real effect there, but unlike some skeptics, my big question with anecdotal evidence is not the well documented placebo effect, but rather that I wonder if there may be confounding variables, even including such things as natural biorhythms.  Again, to be clear, I'm absolutely not simply dismissing the reality of your effects.

May I suggest that you keep daily objective measurements of your performance (and endurance) capacities as in John Mcgough's posts above?  I suppose that a daily subjective record of joint pain wouldn't hurt either.

Staffan Olsson Do you open up every capsule and mix it with olive oil? I have tried that at 20 mg/kg using Swansons and felt no difference.

    • Dean W. Yes, I open up every capsule and mix the fisetin with oil. I think the effect might depend on each person's individual burden of senescent cells.

    • Brian Valerie The effects I got the two years before rapamycin were remarkable. I still get effects from intermittent treatment with large doses of fisetin, but not as pronounced as before

I think it comes down to each persons' individual biology. I have found that since I started with rapamycin, I don't get as strong effect as I did before.

Staffan Olsson Thanks for the info. What time of day to you take the fisetin/olive oil mixture? With or without food? I am just curious to see what might make a difference. I am 74 so I expect to have some senecent cells although I try to stay in good shape, have low cholesterol, average BP, take supplements, etc.

    • Brian Valerie interestingly, it’s been a week since I finish my three days of 3 g per day and I swam again today and I couldn’t believe it but I was again one minute faster. So since I did this protocol, I have improved my 20 lap swim time by 3 1/2 minutes. I had no other explanation as that’s the only thing different in my lifestyle and I have been swimming the same routine for many months.

and I do not open each capsule and mix with oil. I ate some cheese with it and had half tablespoon of coconut oil.

Though I would like to get euphoria reported by Brian, I think it was.

The second day that I took it I also took it with grapefruit juice, and 7 mg of rapamycin.

John Mcgough That really is intriguing, John, and similar to Steffan Olsson's report.  By the way, Stefan is the one who reported "a minor" euphoria, not me.  I do hope that you will continue to keep objective records every day of your performance and endurance to see if this improvement is sustained, as well as to see if there is another improvement after your next three grams for three days fisetin protocol.  Of course, a skeptic might suspect that you subconsciously exerted more effort after the fisetin protocol, but I personally would rather give your quite believable report a tentative benefit of the doubt.  A real correlation in this case is certainly plausible.

    • Brian Valerie it should be noted that the first day I swam while doing the protocol I wasn’t even been thinking about a better performance, and was actually disappointed that I didn’t feel any different on the second day of the protocol.  I was surprised I improved by one minute.  Previously, I had tried to go faster and was not very successful. I had accepted that this was my normal swimming time for 20 laps. And the next time I swam, I was just wondering if my increased speed would hold up and was shocked that it was another minute faster. Physically, I didn’t feel any different going faster as I wasn’t more tired, or had less breath etc.,

it’s difficult for me to believe I can improve that much from one day to the next as I never experience that dramatic improvement before and I had been swimming since March on a regular basis.

Staffan Olsson  Thank you for sharing your info with us! Just a few more questions. Because fisetin is not very soluble I am wondering how much olive oil you use to dissolve or mix your 1 day dose.

And the other questions are about your rapamycin use. How much do you take and how often? Also where do you get it? My doc refuses to prescribe it because she knows nothing about it. This is true of too many doctors!

      Thanks.

I mix the fisetin in a shot, 3-4 cl, of oil (Olive or canola).

I get the Pfizer's rapamune prescribed by a doctor. ( if you live in Europe, that is a potential option in a few countries).

When it comes to dosing for longevity, we all fly in the dark with navigating by the stars. Now I am 6 mg once a week, considering 10 mg every 10 days.  I know that some people take 1 mg every third day. The fact is that no one know the optional dose for longevity.

This calculator can be used to se how rapamycin is metabolized in an average person.

    • Dean W. you did not ask me, but I will add my information. I take my dose with cheese and half a tablespoon of coconut oil and do not mix it.
in regards to Rapa I take 6 mg the first week 7 mg second week 9 mg the third week all with cheese and 8oz. grapefruit juice.

Blagosklonny says the best dose is the largest one you can take without side effects. I also checked my neutrophilous periodically, and they have never been affected by my dose.

The reason I take one large, 9 mg dose is Blagosklonny says a large dose is necessary to pass through the blood brain barrier.

  • Just ordered some.  We'll see what it does to someone's who's pushing 70.  I understand that the right dose is 20 milligrams per kilogram.  At 190lbs that's 17 one hundred milligram tablets.

Well yea. I guess start with that much and see. I started with 18 and now I do 30. There are no side effects and the dosage is not noticeably different at all. Fisetin seems to be quite safe.

let us know if it has an effect. I’d like some backup on this. I swear by this stuff but it’s always nice to see other people say the same.

I'm not going gentle into that good night.   I'm fasting now from dinner to lunch and sometimes a little longer.  I've been trying to figure out when the effect of autophagy take place as I understand the first things the body chows down on in senescent cells.   That almost too good to believe.  But I can't find any consensus on how long the fast needs to be or if shorter intermittent fasting can work.

Robert Olin For fasting I hear it's at least two days, this was from David Sinclair.

    • Robert Olin you should read the research on the fast mimicking diet. I find it very impressive and you are allowed to eat 800 cal per day and supposedly this  produces an even better effect then water fasting.

I have personally done it about 16 times. I don’t buy their food as it is not difficult to imitate their protocol.

whenever I did it, I noticed my lymphocytes jumped up over 100 points.

people with MS do it each month for five days as they claim it regrows the sheath on your spinal canal.

I have also done 3 g of fisetin for three days with no ill affects  and greatly increased my exercise capacity.
