Fisetin experimentation one year (reposted as topic for info purposes)

Hello everyone I wanted to make my first post here as I have been doing a fisetin routine for a year now just about and I have results that are not placebo and very profound. I honestly do not know if this is the senylotic effect or some kind of interaction with fisetin in other ways. 

 

To get it out of the way, I will give the fast and short version. I am taking Swanson’s Fisetin. I take around 2g a day for 3 days( now 2.5g). I repeat this for 3 or 4 times with varying intervals (I was trying to find optimal intervals).  

 

My my first regimen was 3 days treatment with a 2 week delay, then a 3 week delay, then a 4 week delay, then a 2 month delay. I am now convinced a 3 week delay is the minimal amount of time you want taking this the way I am, though I will continue with 4 week delay for safety sake and because of my own observations. 

 

So results. Well I feel like this must play on hormone levels in the short term. Immediately after starting a 3 day regimen, usually on day 2 or 3 I feel strong as hell. In particular my legs are tight and super strong. My muscles in general are firmer and overall strength is amazing. My mind is clear, thinking concise anddynamic. I feel more dominant, self confident, and aggressive. Libido is enhanced as well. This all led me to believe that this acts as a testosterone mimic or elevates testosterone in the blood which is why I cycle monthly and then take a few months off every so often. 

 

i can tell you that at the age of 43 this makes me legitimately remember what early 20s feels like. The effects of one 3 day regimen last strongly for 2 weeks and even on for 4 weeks. In particular during week one and two I was amazed at the amount of pure emotion and clarity of the world around me. I could hear and appreciate sounds and colors seemed more vibrant. Mentally I am on overdrive with super memory and crystal clear logical thinking. I remember the first time I took this I was shocked. I was in a cafe just looking at a coke I had bought in a glass amazed at how more real and present it looked. A small thing but it was profound just how the world seemed more alive. It literally felt like going back in time 20 years. 

 

So for long term effects. A stronger posture, better muscle tone, improved gum health, vastly more energy, in fact this has become all the norm after around 8 months of this. Many of the effects I described in the first two weeks following a 3 day regimen do taper off to an extent, but never completely. 

 

As far as side effects there has not been anything thus far. When you are taking the 3 day dose you might get amped up a little jittery but that gives way quickly after the last dosage and it normalizes into a even effect that is positive in nature. I was very concerned about hair loss as I could not believe how impactful this was. But I have noticed no hair loss at all. I cycle it with a 3 or 4 week break to ensure that there is no negative effect on hair or otherwise. With a two week interval I had some scalp itchiness. That was enough to back me off to to four week intervals.

 

I honestly think that most of what I experienced is not the senylotic effect. It’s way too fast and profound. But I have no ill effects and feel incredible. And overall I have a youthful vigor and look and feel younger. I think a senylotic evaluation, as far as a laymen just observing himself goes ,can only really be done after 2 years or so. However the initial results ( especially gum health) and from what else I can observe are overwhelmingly positive. 

 

I can tell you that this does have an affect and it is profound and noticeable. How much of this is hormone I do not know. But I have other improved health as well, however it’s always tricky to quantify this objectively yourself sometimes. 

 

Good luck on your experiments. Let me know if this has the same effect for you. I just bought Swanson’s with nouvestein. Nothing fancy. Cheap and on amazon.

  • Anyone have any info on a peptide called FOXO4-DRI that is supposed to suppress FOXO4 and increase senescent cell apoptosis?   Is it available?

Well that was fast, yesterday you didnt know much about the substance and now less than a day later you already ordered some. I am impressed with your level of risk tolerance ordering from some random supplier with no assurance of quality or safety and is labeled not for human consumption and you are ready to start injecting it? So what dose are you going to take?

    • Fred Cloud Do you drive down the street with a mask on?   Risk tolerance goes up when you are old and in pain.   But it's MY choice.   Not yours.    The Bill Gates of the world want to vaxx you and make Billions.   They have no incentive for you to live longer and muck up their world.

    • Robert Olin Of course it is your choice, did I tell you that you shouldn't use it? I only  see that I gave you a compliment for going for it. I usually take a few months of research before I feel comfortable taking something risky and people still criticize me for being careless, so I have always considered myself having high risk tolerance more so than most people. But I never attack them, especially if they give me a compliment for going for it.

  • Guys, I have a largely theoretical topic/concern. Could there be any damage to healthy cells from the fisetin?

     

    Logically thinking, if it is an anti-proliferative agent, invoking apoptosis, could it non-selectively damage healthy cells also? Like the chemotherapy, which would also damage healthy cells.

     

    Another similar concern is about it being topoisomerase inhibitor, how bad could it really be for the body?

     

    And from the wikipedia, what do they mean by this?

    • Andrew Whitfield Other senolytics like foxo4-dri definitely do have collateral damage and kill off healthy cells, so your theory has merit. So for every 10 senescent cells foxo4-dri kills off it also kills off one healthy cell which is alot and is why one of the lead researchers said he felt foxo4-dri isn't safe. But fisetin seems to have more selectivity and so I think you are safe. Having said that I wouldn't take anymore than you need to. Some people take 2-3 grams per dose and have reported issue with healing and bleeding. Some of the more conservative doses scaled up from mouse studies say 500mg should be effective. If you take a round and can feel some of the effects of killing off senescent cells, alleviation of some chronic condition or even just post round hunger might be sufficient to dial in the dose to a minimum effective dose.

  • Unlike chemo drugs that directly target and kill cells, fisetin does not do that. What it does is that it induces cell death. In other words, it encourages the body to remove senescent cells. I hope that you can appreciate the difference.

      Thanks. But how does it choose selectively? Is there a risk of harming healthy ones? I have read some descriptions of dropped WBC in blood test after it. Which sounds a bit alarming.

       

      And what about the other point I mentioned, like inhibition of topoisomerase?

      Andrew Whitfield Well the truth is the mechanisms behind fisetin are not that well known. There are good theories but i have read different ones. The main thing to remember is that they fed mice huge doses and nothing bad happened but increased health and life span. 
       

      So most of this stuff is take at your own risk. However Fisetin seems fine to me. Over a year on it. Even FOX04 is targeting a specific pathway. There is no guarantee that somewhere somehow good cells are not being affected negatively. 
       

      Any senolytic you take in these early days carries some risk as we need decades of observation to truly understand how these things work in humans. The thing is do you have decades?

  • Andrew Whitfield Fisetin does not "choose selectively" to destroy senescent cells. So why does taking fisetin remove senescent cells? The simple answer is that your own  genes remove senescent cells. Fisetin "triggers", "induce", "causes" those genes to activate, when they receive chemical signals from fisetin.

    Your body will not get signals from senolytics and start destroying perfectly healthy cells.

    The danger is, fisetin is or at least might be toxic at very high levels. So, you look at the safe dosages and how long to take it, the recommended course of taking fisetin.

    • Andrew P K Yap Fisetin (and Quercetin) inhibit the expression and activation of the PI3K/Akt/mTOR signaling pathway in some types of cells which enhances apoptosis. In some studies it was observed to suppress proliferation of tumor cells through this mechanism. Sheeky shared a rather nice diagram of different senolytic pathways this week;

  • Sorry for any grumpiness.    My backs been aching 24/7.  Had some endoscopic surgery 1.5 years ago and I'd like to avoid doing it again.   Yes taking FOXO4-DRI is a risk but so is rock climbing.   I do appreciate your feedback but like Dylan said "When you ain't got nothin you got nothin to lose."

      Robert Olin There have been self experimenters who had no issues with it. The guy at buckylabs if I am not mistaken took it for a long time.

    • Robert Olin No worries Robert, hope your back heals.  Just off the top of my head I would think that for back pain you would want a senolytic that targets fibroblasts like azithromycin but you may want to research it to confirm.

    So as I have mentioned earlier, I am on bisoprolol and perindopril for my hypertension.

    Ive just got a Swanson Fisetin 100mg pills, and have tried two dosings, 1 pill and 2 pills in two evenings before sleep. I did not notice any ill effects, and my heart is also fine (HR and BP on usual levels).

    I will follow an unusual (safer) approach doubling the dose until I reach 1200-1400. So today I will try to dissolve 4 capsules content in olive oil and take it in the evening.

     

    Will report more later on! :)

  • I have tried putting the 4 capsules content into extra virgin olive oil. It was absolutely disgusting! A bitter taste, powder did not dissolve well.. Guys, how do you resolve it? Are you choking on 10-15 pills, ingest pure powder or use some other way. Please share some advice.

      Last year I ordered 100 gm of >98% pure powdered Fisetin from VitaSpace for $190 plus $6 shipping.   I put Trader Joe's Extra Virgin Olive Oil in a shot glass, heat it the microwave for 40-60 seconds until its quite warm but drinkable, put the shot glass on a milligram-sensitive digital scale that is tared to zero, add 2 grams of the Fisetin powder, stir vigorously until I get a mustard-colored liquid suspension (and possibly partial dissolution) of the Fisetin powder, and then gulp it down with a toss.  Then I scoop out the residue with bread fragments and eat those.  The taste isn't great, but I wouldn't say it's disgusting.

    • Andrew Whitfield try buying a mini blender.  I bought a tribest off amazon.  2-3 cups small. I blend all my pills to get them down.
