I was wondering how you made your cream the 2mg in 4oz? 4oz of what?  Thank you.

  • From the other thread: Ok I am taking Arbo Trex Dmso/Aloe Vera cream.... 70/30 with Lavender scent mixed DMSO containing 3 mgs of Sirolimus mixed in Liquid DMSO.  I use a mortar and pistol to grind down the pills and then put them in a text tube with DMSO to thoroughly mix them up. Mixture will be Cloudy with dissolved Sirolimus. Then I mix it into the 4 oz cream. You can find the Trex cream and pure DMSO on Amazon.  I am awaiting my Rapa and have already purchased these for the solution and mixing, the percent Rapa is 0.0026%, tech papers have indicated 0.001-0.015 range with results in 90-180 days depending on the ailment or improvement and how many applications per day, up to 2 max.

    • Steve Olsen I started that exact regimen on 11/5, with 2 applications/day. On 11/18, I had the strong impression that my wrinkles had gotten worse. I began to wonder if mTORC2 was being triggered, so I reduced to 1 application per day and took an occasional day off. On 12/13 visible wrinkles and skin fold test appeared the same or worse again. None of these observations were rigorous, but I'm fairly confident that there has been no improvement after a month and a half.

      Besides Mark, has anyone had measurable or even the impression of significant improvements, and if so what did you do and over what period of time?

      If activation of mTORC2 is a concern, as it is with internal rapamycin, how do we avoid that while maximizing activation of mTORC1?

    • chuck stanley  Sorry to hear your results were poor. On my face the dmso was to tough so I had to stop. My arms and hands are doing well, once per day though. I made up another creme with the same weight percent Rapa and it is doing fine on the face, but the DMSO really drives the Rapa in the skin and seems to work quicker. Being we are early adopters we will run into issues and have to work through them. I suspect the dmso formula drives Rapa into our blood, so have been using only 3 to 5mg once per 7 days internally. Too soon to tell anything , hands and arms seem to be improving, face is healed and hopefully will improve as well. Studies took 6 to 8 months, but lower dosages. Skin turnover will also limit things probably every 30 days or so per rep. Taking pics for evidence.

    • Steve Olsen Michael Thanks for the feedback. After my disappointment with twice a day application of topical sirolimus, I then tried several weeks of once a day application and still saw no positive results. But knowing that many followers are applying  twice a day, and considering how much higher the concentrations are that are being used to treat skin conditions I figured it wasnt likely that the small amount I was using (~1/4 teaspoon total for upper face above beard line plus for lower neck) probably wouldnt be enough to activate mTOR2. So I relented and went back to 2 applications per day, at least on my face. After a week or so I by chance noticed that the finest lines on my face had become hardly visible. These were the tiny, very fine stretch marks from sagging of my cheek to the marionette lines.  In retrospect I believe my hopes for seeing something dramatic led me to not notice what was happening more gradually. So I am encouraged by those results, now keeping my eyes on the tiniest lines rather than the deeper wrinkles and trying to be more patient.

    Just received my 3mg capsules of rapamycin for topical purposes.  Dr. Green recommends mixing in 3.5oz of Aquaphor Healing Ointment.  But my experimental mind thought first of the aloe / DMSO combination as a potential superior solution.  And a google search brought me here.  So I'm very interested to hear of any further results or feedback from those who have tried this.  Thank you.

    • Andrew you could search the forum for DMSO/sirolimus to find Mark Thimineur's post for his suggested mixture.

      After 5 months I am convinced that it works on fine wrinkles and elasticity, which is noticeable on the back of my hands. It hasnt kept up with the wrinkling that occurs as loss of volume in my  face. That has gotten worse. But then again, I'm over 80.
