Rapamycin - progress so far

Hi all, 

About me: I'm 46 y.o. medical doctor based in Europe and started taking weekly rapamycin 4 months ago. I go to the gym regularly and am in average physical condition for a guy of my age. The last couple of years have brought the first negative signs of physical aging: aches and pains that don't go away, increasing belly fat, a decline in gym performance, wrinkles and increased pigmentation, grey hairs as well as slower wound healing. Am venturing on this rapamycin journey to see whether it can  slow down the inevitable age-related decline and see how it impacts my aging longer term.

Where I'm at now: I started rapamycin 4 months ago at 2mg once weekly and have been slowly increasing the weekly dose by 1mg each month. Now at 5mg per week. I intend to continue on this dose increase scheme until I reach the highest optimal dose, beyond which side effects become an issue.

Currently have experienced zero side effects, not even mouth ulcers.  I have noticed improvements already in body composition (slight reduction in body fat) starting at the 3mg/week dose and most recently increased gym performance (cardio vascular and muscle strength increase) since being on 5 mg/week.

Intend to progress to 6mg/week in September.

Will at some stage probably try topical rapamycin on wrinkle / pigmentation problem areas as well. In Europe a rapamycin solution (Pfizer) is easily available in pharmacies.

    Rapamycin can reduce body fat, but there are more suitable means for this purpose.

    I’ve spent a lot of time training, but even after lots of practice, I couldn’t see a pleasant result for me. I didn’t know what was the problem until I figure it out. The biggest problem for me was the unsuitable fitness machine. Especially I couldn’t understand why the calves were so hard to gain. Therefore I advised the gym to change the calf raise machine.     • Van I would really like to see more self experimentation with Fisetin at 2grams a day or more.

  • Diamond Tree  hi I am also in the Western EU, where did you get the prescription from? I have tried many doctors but they refuse to provide a prescription unless I am getting organ transplants.

      If you have a friend in Turkey, he can send you Rapamune. I am from Bulgaria and here you can not do without a prescription, and they send it to me from there. Another option is from Ukraine or India ... Sorry for my bad English.

    Hi George, I was wondering if you’ve had any problems with skin infections (which would indicate a weakened innate immune system).  I’m 59 y/o and have been taking 6mg/week over past 9 months and gotten 2 infections that seemed to proceed quite quickly. Treated both with antibiotics successfully, but am concerned that I’m weakening my immune system too much (too much MTOR inhibition?).  I’m going to start taking 6mg every 2 weeks to allow the rapa to fully exit my system.  Also seems logical that we should fully alternate between low MTOR and normal MTOR levels. 
     

    What might also be complicating my situation is that I take other things that reduce MTOR (eg metformin, curcumin extract, etc).  I’m not diabetic, but take the above for life extension purposes.

    Any thoughts given your experience?

    • Armando in case you dont know about the effect of curcumin on rapamycin, you might want to research it. 

      I take an estimated dose of 21 mg biweekly. No rashes. I started out with 6 mg weekly and gradually built up to the dose I now take.

  • Anyone  here dealing with skin rashes from rapamycin? I get skin rashes from just 2 mg weekly, about every second time or so. If untreated they get unbearable; with anti-inflammatory cream the rashes are quickly subdued. Either I will be hyper-vigilant just needing a small amount of anti-inflammatory as soon as the first itching starts, or just take the 2 mg every 10 days or so.    Seems no way I can do the high doses mentioned on this thread. Maybe try 4 mg every two weeks......  trying to find my way.

    • Ulf Rasmusson mild rashes are listed as a common side effect of rapamycin. I get itchy but no rashes.

    • Ulf Rasmusson I’m running into the same problem with skin rashes/bumps. I just started the 2mg dose once weekly and in a matter of a few weeks, skin and UR infections started. What did you use to treat your skin, please?

      Maria Molè Thank you. I also restarted exogenous testosterone. That must be the culprit.

    • Maria Molè Just ask for an anti-inflammatory cream at any pharmacy, nonprescription. Away from home, so  can't give you any brand name but I don't think it matters which.

  • I take 15 mg of rapamycin every two weeks.

    Recently had epigenetic age measured by TruDiagnostic. Was about 9 years younger than chronological age which agreed with Levine calculator

    One unexpected report was on what they called Extrinsic age which was described as bio age of immune system. For me the report showed a 19 year reduction in my immune system age.

    No sign that rapa is having a negative effect on my immunity.

  • Has anybody noticed or heard about a sharp drop in neutrophils after taking rapamycin? When I upped the dose to around 6mg bi-weekly (2 mg + fresh-squeezed citrus juice) my neutrophils dropped 50 %. No change in lymphocytes or red blood cells.

    When I reduced to around 3mg bi-weekly, neutrophils only dropped slightly.

    Will try papaya fruit extract to bump up my neutrophils. If this doesn't work it seems I have to stay far from the dose I would like.

    There ain't  no free lunch.....

    • Ulf Rasmusson Incredibly, high dose papaya fruit extract boosted my neutrophils 80 % in three weeks. Can resume rapamycin, whilst continuing with the extract  which upped my lymphocytes as well, by 40 %.

    • Ulf Rasmusson My neutraphil % went up but my lymph% came down, dropping below normal range with rapa so my doctor recommended a mushroom formula.

      It will be some months before I know if it worked.

    • chuck stanley Could you share the mushroom formula? After the papaya, my neutrophils are in the ok range, but lymphocytes are still way below the normal range after getting drastically hit by dasatinib in D+Q
